1. UV spectrum of lutein and its lutein ester

1.1 UV spectrum scan of lutein ester extracted with acetone after reference to lutein reference substance and methanol treated marigold, such as Fig.1 and Fig .2.

Fig.1 UV spectra of lutein

Fig .2 UV spectra of lutein ester

From Fig.1 and Fig.2 shows, Both have the same characteristic spectrum, the maximum absorption peak wavelength in acetone is 446 nm.

1.2 UV spectrum of methanol, ethanol and isopropanol extracts

UV spectra of extracting solvent with methanol ,ethanol and isopropanol from marigold such as Fig.3

Fig.3 UV spectra of extracting solvent with methanol ,

ethanol and isopropanol

From Fig.3 shows the solubility of lutein ester in methanol, ethanol, isopropanol decreases with the polarity of the alcohol, dissolves

Gradually increasing. It is a substance with high alcohol solubility at 370 nm, and its main components are syringic acid, quercetin, etc. Since the solubility of this substance in methanol is large and the solubility of lutein ester in methanol is small, so we chose

1.3 Drawing of standard curve

HPLC spectra of lutein such as : Fig.4

1.4 Selection of extraction conditions of Lutein ester

Marigold flowers are pretreated with alcohol and treated without alcohol, which has a great influence on the extraction rate of lutein ester. The highly polar substance in the flower hindered the dissolution of lutein ester by N-hexane. The comparison results of the methanol-treated marigold flowers and the alcohol-free extraction rate are shown in Fig. 5.

Experimental summary

Pretreatment of marigold flowers with methanol not only greatly accelerates the extraction rate of lutein ester, but also obtains a higher purity of the ester, which is very advantageous for the later saponification. This method is not only suitable for the extraction of flowers, but also the extraction of dried flowers.
